Hello Julie,
I will take half a day to reach Nusa Penida from Pujut. Because you need to reach Sengigi or Bangsal and then take a fast boat to Nusa Penida. So the only way to go diving in Nusa Penida would be to stay one or two nights in Nusa Penida. If you are interested I sent you by email the contact of my friend Roy who can organize the full trip including the diving day in Nusa Penida.
Alternatively you may consider to dive in Lombok.
